Synergistic lethal effect between PI3K/mTOR inhibitor Voxtalisib and arsenic trioxide on human leukemic KG-1 cell line

Abstract: Myeloid/lymphoid neoplasms with FGFR1 rearrangement(also known as 8p11 myeloproliferative syndrome, EMS) is a distinct disease entity in the current WHO classification. These patients frequently (~80%) progress to acute myeloid leukemia (AML) and have a poor outcome with a 5-year survival rate of < 20%. To identify the potential treatment regimen on these patients, the four reagents (Idarubicin, arsenic trioxide, Voxtalisib and Ruxolitinib) were applied on KG-1 cells. Our results showed that the KG-1 cell line was resistant to  idarubicin. However, the dual PI3K/mTOR inhibitor Voxtalisib, rather than JAK1/2 inhibitor Ruxolitinib could be effectively against KG-1 cells. Arsenic trioxide(As2O3) combined with Voxtalisib synergistically inhibitted the viability of KG-1 cells. We also found that the combination treatment more significantly reduced the colony formation, induced apoptosis, decreased Bcl-2 expression, but increased caspase-3 expression as compared to the single drug treatment. Additionally, As2O3 enhanced the effect of Voxtalisib which decreased the phosphorylation of PI3K, AKT and mTOR. These data suggests that Voxtalisib in combination with As2O3 may provide a novel and efficacious therapy regimen for patients with EMS.

Effects of selenium and silicon fertilizer on enhancing resistance of flue-cured tobacco

Abstract: In order to evaluate the role selenium and silicon fertilizer in enhancing resistance of flue-cured tobacco, flue-cured tobacco variety Zhongyan 100 was potted. The effects of selenium and silicon fertilizer were evaluated by analyzing the carbon and nitrogen metabolism, antioxidase and resistance index of flue-cured tobacco. The results showed that selenium and silicon fertilizer could promote the expression of key enzyme genes in carbon and nitrogen metabolism, including Sucrose phosphate synthetase (SPS), sucrose synthetase (SS), soluble starch synthase (SSS), nitrate reductase (NR), glutamine synthetase (GS) and glutamate dehydrogenase (GDH), and increase the activities of resistance-related enzymes Superoxide dismutase (SOD), Peroxidase (POD), Catalase (CAT) and Ascorbate peroxidase (APX). Both selenium and silicon fertilizer can increase the contents of NPT, ASA and GSH, and reduce the content of hydrogen peroxide (H2O2) and malondialdehyde (MDA) in flue-cured tobacco. The effect of selenium fertilizer is more obvious than silicon fertilizer. These results indicate that selenium and silicon fertilizer can promote the formation of healthy tobacco plant through enhancing carbon and nitrogen metabolism and improving the regulation ability of redox balance in flue-cured tobacco.

Characterization of plant spacing best fit for economic yield, fiber quality, whitefly and CLCuV disease management on upland cotton

Abstract: In order to find out best fit plant spacing for upland cotton an experiment was conducted at cotton Research Institute, Faisalabad during 2012. Treatments were four spacing (plant to plant and row to row) with 2 varieties. From the results it was found that traits like days taken to first bud, first flower, number of bolls, number of monopodial as well as sympodial branches and boll weight directly contributed toward economic yield of seed cotton. Economic yield of tested varieties i.e., FH-114 possessed more yield (1571.5 kg/ha) as compared with FH-4243 (1200.0 kg/ha). Based on the results of interaction between varieties and spacing, economic yield of both varieties was improved in 75x15cm combination (1492.7 Kg/ha), however yield showed declining trend under high population/dense spacing 37.5x15 cm (1282.1 kg/ha). Dense spacing declined the economic yield (1282.1 kg/ha), fiber quality (5.1µg/inch), but promoted whitefly (9.0/leaf) and cotton leaf curl disease incidence (81.6%) as compared with 4.7/leaf whitefly and 55.4% disease incidence in 75x30cm spacing combination. Incidence of whitefly and disease successively increased with dense spacing and plant populations. From the results it was concluded that proper plant and row spacing is very much important for increasing yield and productivity of cotton, however bet fit plant spacing for FH-114 and FH-4243 was 75x15cm combination for row and plant spacing respectively.

Serum and urinary NGAL and Cystatin C levels as biomarker tools for diagnosis of both AKI and CKD: A histobiochemical comparative study

Abstract: Introduction: Acute kidney injury (AKI) has a high global incidence with observable complications in critically ill patients. Long-term disease and medication complexity contribute to devastating chronic kidney disease (CKD) and diminish quality of life. Establishing new biomarkers would guide patient care and facilitate novel therapeutics development. Methods: Serum and urinary levels of creatinine, CysC, and NGAL were estimated in 86 renal patients and compared with healthy controls for AKI and CKD categorization. Creatinine and Cystatin C (CysC) measurements were used to estimate GFR. Biopsy tissues were prepared for light microscopy for further characterization. Patients’ demographic data were used in group association study. Results: Thirty-six patients met the criteria for AKI and 50 for CKD. Mean values of serum CysC were higher than controls but similar in both disease states, while urine levels were slightly high in CKD patients and remained steady by the end of follow-up (EF-Up). Further, a 2.7-fold and 5.5-fold increase in serum NGAL were observed in AKI and CKD, respectively, and a dramatic 7.0-fold reduction in AKI group at EF-Up. Similarly, urine NGAL for AKI and CKD increased 3-fold and 6-fold respectively on admission, which multiplied to 7.3-fold and 10.7-fold at EF-Up. ROC assessment curve revealed a relatively higher NGAL performance at good predictive values than CysC (p< 0.009). Conclusions: We showed a higher urine NGAL and CysC sensitivity and specificity than their serum counterparts, providing a powerful discriminative tool between AKI and CKD. CysC, however, displayed less sensitive performance than NGAL, indicating effects by enigmatic non-specific factors.

Abstract: From the Holy Bible we have an interesting info. that Adam lived for nine hundred and thirty years , the days of Seth were nine hundred and twelve years, Enoch’s life time was nine hundred and five years, , all the days of Cainan were nine hundred and ten years, and Mahalalel lived for eight hundred and ninety five years, [Genesis,5:5, 5:8, 5:11, 5:14 and 5:17 ] In those period, there were no pollution. In those days, there were no chemicals and fertilizers for farming. The air was pure, the water was clean. The nature was in favor of good health. But, now a days the scene is entirely different. To live for hundreds of years without any disease, a new natural medicine has been introduced.

ABSTRACT: In presence of high levels of blood LDL particles, free radicals (ROS), macrophages, cytokines, platelets, histamine, 5HT get involved in LDL oxidation, leading to formation of atherosclerotic plaques which get deposited in endothelium of coronary arteries causing coronary artery disease (CAD). Psyllium fibers and Ficus Carica (FC) are being considered to be useful in treating hyperlipidemia, hypertension, and hyperglycemia. This study was conducted to determine hypotensive, hypolipidemic, and hypoglycemic potential of Psyllium husk and FC. Study was conducted in National hospital, Lahore, Pakistan from January to April 2019. Seventy five hyperlipidemic, hypertensive and diabetic patients were enrolled selected from the hospital with their consent. They were divided in three groups ie 25 patients in each group. Group-1 was on placebo, group-2 was on 15 grams psyllium husk to be used in three divided doses. Group-3 was on 12 grams FC to be taken in three divided doses daily for two months. Their Lipid profile, FBS, and hypertension were determined before and after two months therapy by these two agents. Two analyze data we used SPSS version 10.01.001 2015 and t test was applied to determine changes in the tested parameters. In two months therapy by psyllium husk in 24 patients TG, TC, LDL-c was reduced 21.48, 10.09, 24.17 mg/dl respectively. HDL was increased 12.38 mg/dl in this group. FBS reduction was 28.66 mg/dl, systolic BP 6.30, and diastolic BP reduced in this group 7.52 mm of mg. We then concluded that FC and PH have good enough potential to normalize lipid profile, reduce BP, and FBS in patients suffering from metabolic syndrome.

ABSTRACT: To treat metabolic syndrome by allopathic drugs cause side effects so Herbal medicines are replacing drug treatment of hyperglycemia, hypertension and hyperlipidemia especially by antioxidant effects of their active ingredients.  We have compared antioxident characteristics of vitamin-E and red Dates. It was placebo-controlled conducted study conducted at General Hospital Lahore Pakistan from January 2022 to march 2022. Inclusion and exclusion criteria: 120 male hyperlipidemic patients were included with age range from 18 to 70 years. We excluded patients already suffering from renal, hepatic, pulmonary, or thyroid diseases. We also excluded patients already on cardio and hepato-protective drugs. Sample size and division of patients: 120 hyperlipidemic patients were divided in three equal number of patients, 40 patients in each group. Group-I (n=40) was on placebo. They were provided capsules which were filled with grounded brown rice taking 8 hourly daily for one month. Group-II (n=40) was on capsules of vitamin E 400 mg taking eight hourly daily for one month. Group-II (n=40) was advised to take red dates 20 grams thrice daily for the period of one month. Method of lipid profile determination: LDL-cholesterol was calculated as total cholesterol- HDL-cholesterol-VLDL-cholesterol;VLDL-cholesterol was measured directly after ultracentrifugation. Observations and Results: After one month therapy their post treatment lipid profile was determined and analyzed statistically by using SPSS version 2.2.01 2013. Paired ‘t’ test was applied for mean values with SD of the parameters before and after treatment. Results showed highly significant change in LDL-cholesterol of group-II patients but HDL-cholesterol was increased 6.6 mg/dl, still it was significant change with p-value of <0.01. In group-III LDL-cholesterol was decreased 10.9 mg/dl which is significant change with p-value <0.01. HDL-cholesterol in this group was increased 4.2 mg/dl which is non significant with p-value of >0.05.  Conclusion: We concluded from the study that Z. Jujube has antioxidant potential by lowering LDL-cholesterol in human plasma. But This effect is not comparable with hypolipidemic effects of Vitamin E as it also increases good cholesterol i.e. HDL-cholesterol.
